Gonadotropin releasing hormone-induced histone modifications and gonadotropin gene expression

Melamed Philippa , Wijeweera Andrea

Gonadotropin releasing hormone actively de-represses expression of the gonadotropin subunit genes through several actions that target the chromatin. We have shown in the past that both LHβ and FSHβ genes are repressed by histone deacetylases (HDACs) in αT3-1 gonadotrope cells and that this repression is overcome by exposure to GnRH which facilitates HDAC removal via CaMKI-mediated phosphorylation. At the LHβ gene promoter, both histone acetylation and phosp...

Hyperglycemia causes a reduction in FSH levels following aberrant epigenetic regulation in the gonadotropes due to the increased glucose metabolism

Melamed Philippa , Feldman Alona , Saleh Ayah , Pnueli Lilach

The connection between metabolic state and fertility is well-recognized, and the hypothalamus clearly plays a central role in translating metabolic signals into altered GnRH release which affects the reproductive axis. We hypothesized that the gonadotropes might also directly impart some of the effects of hyperglycemia on reproductive function. Gonadotropes were shown to express predominantly the insulin-independent Glut-1 transporter, and their incubation in high glucose incr...
